Pengembangan Nematoda Entomopatogen Sebagai Agens Pengendali Hayati Dan Uji Patogenesitas Terhadap Larva Spodoptera litura
Abstract
Entomopathogenic nematodes are one of the biological control agents of pests on plants by utilizing natural enemies of pests. Entomopathogenic nematodes are used as another alternative to reduce the use of chemical insecticides. The purpose of this study was to determine the value of Lethal Concentration (LC50) and population density of entomopathogenic nematodes that were effective in killing S. litura larvae. The methods used are in vivo and in vitro. The experimental design used was a completely randomized design (CRD) using five treatments with different concentrations of NEP (P0 (Control) = 0 IJ/ml, P1= 200 IJ/ml, P2= 400 IJ/ml, P3= 600 IJ /ml, P4 = 800 IJ/ml) each treatment was repeated 4 times. Parameters observed were larval mortality at 12 hours, 24 hours, 36 hours, 48 hours, 72 hours, 96 hours, 120 hours after application. While the data were analyzed by using SPSS 16.0 (significant degree 0.05%) and Lethal Concentration with probit analysis test. The results of the identification of sugarcane soil samples in Mojoagung District, Jombang Regency, obtained nematodes of the genus Steinernema. The mortality result of S. litura in treatment P2 = 400 IJ/ml showed the highest last observation (120 hours) compared to other treatments, namely 86.67%. The results of the NEP pathogenicity test showed that the Lethal Concentration (LC 50) value was 214 IJ/ml. It can be stated that in NEP isolates to kill 50% of S. litura larvae, a concentration of 214 IJ/ml is required.
