Abstract

Plectranthus scutellarioides or iler plant is a medicinal plant that has pharmacological activities such as anti-inflammatory, antioxidant, and antibacterial. Based on the benefits of Iler Plants, it is necessary to have plant propagation through tissue culture techniques with the addition of auxin and cytokinin growth regulators. The purpose of this study was to determine the effect of the combination of growth regulators auxin (IAA and 2,4-D) and cytokinins (BAP) on callus induction of iler plants. This study used a completely randomized design method with 7 treatments with 3 replications. Explants were grown on MS media, sucrose 30 g/L, and additional growth regulators IAA and 2,4-D (0,5; 1; 1,5 ppm) and BAP (0,5; 1; 1,5 ppm). After 5 weeks of planting, callus was subcultured on the same medium as before. After 4 weeks of subculture, harvesting and observation of callus morphology, fresh weight, and dry weight of callus. The results showed that the callus produced was white, greenish white, brownish white to brown with a compact texture. The highest average value in treatment (D) a combination of 1,5 ppm IAA and 0,5 ppm BAP, namely 20,6 HST at the time of callus formation, 0,2909 gram at callus fresh weight, and 0,0316 gram at callus dry weight
